益气活血方对糖尿病大鼠视网膜微血管病变血管内皮生长因子表达的影响

摘　　要：目的:观察益气活血法对糖尿病大鼠视网膜微血管病变血管内皮生长因子(VEGF)表达的影响。方法:用链脲佐菌素(65mg/kg)一次性腹腔注射的方法建立糖尿病大鼠模型,50只SD大鼠随机分成对照组、模型组和益气活血中药组。益气活血中药(芪参益气滴丸浸膏)每日(0.2g·kg-1·d-1)灌胃给药。10个月后处死动物检测血清VEGF,摘除眼球进行眼球切片,视网膜血管消化铺片和免疫组织化学染色观察VEGF。结果:模型组血清的VEGF含量比对照组升高,益气活血中药组的VEGF含量比模型组低,但差异内无统计学意义(P>0.05)。视网膜全层及毛细血管VEGF表达模型组明显高于对照组,益气活血中药组表达明显低于模型组(P<0.01)。结论:早期干预性给与益气活血中药可通过降低视网膜VEGF的表达,延缓DR的发生发展。Objective：To study the therapeutic effect of Yiqi Huoxue therapy on vascular endothelial growth factor（VEGF） in retina of diabetic rats.Methods：The diabetic rat model was set up by a single intraperitoneal injection of streptozotocin（65mg/kg）.Fifty rats were randomly divided into normal group,model group and Yiqi Huoxue medicine group,the normal group and model group were given normal saline by intragastric administration,and Yiqi Huoxue medicine group was given extractum of Qisheng Yiqi Dripping Pills（0.2g·kg^-1·d^-1） by intragastric administration.All rats were killed after 300days.The serum VEGF levels were measured.Immunological histochemistry assay was used to observe the expression of VEGF of eyeball slice for quantitative analysis.SPSS was used for statistical analysis.Results：Ten months later,the levels of VEGF in serum of model group were higher than those in control group,the levels of VEGF in serum of Yiqi Huoxue medicine group were lower than those in model group（P〈0.05）.The model group rats displayed high expression of VEGF.Compared with the model group,the expression of VEGF in Yiqi Huoxue medicine group decreased evidently（P〉0.01）.Conclusion：Yiqi Huoxue medicine can reduce damage of diabetic retinopathy and this effect may be achieved by restraining the expression of VEGF.
