摘  要:目的研究脑源性神经营养因子(BDNF)在人脑胶质瘤内的表达以及他们与胶质瘤病理分级的关系。方法用免疫组织化学亲合素-生物素过氧化物酶复合物法(ABC法)和Western blot检测了5例正常人脑组织和80例胶质瘤样本中BDNF表达水平。结果免疫组化和Western blot结果显示正常人脑和各级胶质瘤组织内均有BDNF表达,阳性反应物主要位于胞浆。BDNF在胶质瘤中的蛋白水平高于正常脑组织(P<0.05),且随胶质瘤病理级别的增加而逐渐增高(P<0.05)。结论胶质瘤中BDNF表达随病理级别增高而增加,提示BDNF在胶质瘤的发生过程中起重要作用。Objective To investigate the expression and significance of brain derived neurotrophie factors (BDNF) in human gliomas and explore the relationship between BDNF and pathological grades of gliomas. Methods Immunohistochemistry and Western blot were applied to evaluate the expression of BDNF in 5 cases of normal brain tissues and 80 glioma samples of different pathological grades. Results The expression of BDNF was observed mainly in the cytoplasm of both normal brain tissues and glioma tissues. Immunohistochemistry and Western blot showed that the expression level of BDNF in gliomas was significantly higher than that of normal brain tissues (P 〈0.05), and the positive expression rate of BDNF gradually increased with the increase of pathological grade of glioma (P 〈 0.05 ). Conclusion The expression level of BDNF is correlated positively with the tumors'pathological grade, which indicates BDNF may play an important role in the tumorigenesis of gliomas.
