摘  要:以5个玉米自交系幼胚为外植体,研究了基因型、预处理时间、继代时间、培养温度以及器皿等因素对受体系统建立的影响。结果表明:5个基因型的外植体在相同条件下均可诱导出愈伤组织,但是不同基因型间胚性愈伤组织的诱导存在显著差异。继代时间对于愈伤组织生长亦有显著影响。此外,不同培养温度以及不同培养器皿等亦是胚性愈伤组织诱导的重要影响因素。实验得出最佳玉米幼胚转基因受体系统为:基因型78599在4℃冷藏预处理1d,用试管25℃环境下诱导培养2次,继代培养3~4次。The immature embryos of five maize inbred-lines (Zea mays L. ) were used as explants to study the effect of genotype, pretreatment and subculture time, culture temperature, culture containers, and other factors on the transgenic receptor system. The results showed that calli could be obtained from the explants of five maize lines under the same conditions. However,there was significant difference among the embryonic callus induction for different genotypes. The subculture time had obvious effect on the growth of the callus as well. Besides,the culture temperature and culture containers were important factors for the embryonic callus induction. It indicated that the best transgenic receptor system for immature embryo of maize line 78599 was that immature embryo was cold-treated for one day and induced twice in test tube,then subcuhured 3 -4 times at 25 ℃.
