摘 要:目的建立不同产地黄柏药材中4种主要生物碱类成分的含量测定方法。方法采用高效液相色谱法,色谱柱为Diamonsil C18(250 mm×4.6 mm,5μm),流动相为乙腈-0.5%三乙胺(梯度洗脱),流速为1.0 mL.min-1,柱温为25℃,检测波长为284 nm,同时测定黄柏药材中黄柏碱、药根碱、巴马汀和小檗碱的含量。结果4种成分均能达到基线分离,线性良好。不同产地黄柏药材中4种主要生物碱类成分的含量差别较大,产自四川泸州黄柏中的黄柏碱、甘肃武都黄柏中的药根碱、广西桂林黄柏中的巴马汀、四川雅安黄柏中的小檗碱含量分别为最高。结论本方法操作简便、结果准确、重现性好,适用于黄柏药材中4种主要生物碱类成分的含量测定。Objective To establish a simultaneous determination method for four main alkaloids in Cortex Phellodendri Chinensis from different habitats by HPLC. Methods HPLC was performed on Diamonsil C18 column(250 mm×4.6 mm,5 μm) with acetonitrile-0.5% triethylamine as mobile phase(gradient elution) at a flow rate of 1.0 mL·min-1.The column temperature was maintained at 25 ℃ and the detective wavelength was set at 284 nm.The contents of phellodendrine,jatrorrhizine,palmatine and berberine were determined simultaneously.Results All the four alkaloids achieved baseline separation with good linearity.The contents of the four main alkaloids in Cortex P.Chinensis from different habitats varied significantly.Respectively,the contents of phellodendrine in Cortex P.Chinensis from Luzhou of Sichuan,jatrorrhizine in those from Wudu of Gansu,palmatine in those from Guilin of Guangxi,and berberine in those from Yaan of Sichuan were highest.Conclusion This method is simple,reliable and repeatable,and is suitable for the determination of main alkaloids in Cortex P.Chinensis.
