观赏植物‘孩儿莲’及其近缘植物差异蛋白组的电泳分析

摘  要:为探讨八角属植物结实性差异的分子机理,以‘孩儿莲’、‘杭州红茴香’及披针叶八角的幼叶为材料,在优化蛋白质双向电泳技术的基础上,就3种供试植物叶片蛋白质组的差异表达进行了分析。结果表明:在采用改良酚提取法、蛋白溶解液含0.25%IPG缓冲液、等点聚焦条件为50 000 V.Hr时,可以得到分离效果良好的双向电泳图谱。经2 DE Image Master软件分析,结果从‘孩儿莲’、‘杭州红茴香’和披针叶八角的电泳图谱上分别检测到6626、44和684个蛋白质斑点,说明在蛋白质分子水平上,‘杭州红茴香’与披针叶八角的亲缘关系较近,而与‘孩儿莲’的遗传差异较大。To investigate the molecular mechanism of fruitfulness difference in Illiciaceae plants, differential proteome were analyzed by using young leaves of Haierlian, Hong hui xiang, Hlicium lanceolatum A. C. Smith on the basis of optimizing 2-DE technology. 2-DE maps with good separation were obtained when IEF condition was 50 000 V · Hr using protein dissolving solution containing IPG buffer concentration 0. 25 % and phenol/SDS buffer for protein extraction. Analyzed by 2-DE Image Master software, 662,684 and 644 protein spots were detected respectively from Haierlian, Hong hui xiang, Illicium lanceolatumA. C. Smith, and showed that Hong hui xiang and Illicium lanceolatum A. C. Smith had close genetic relationship and Haierlian had more genetic difference with them.
