摘 要:通过肠道细菌分离筛选技术,从云南云秃蝗(Yunnanacris yunnaneus)活体标本肠道内分离出可培养细菌22株,并对22株菌基因组16SrDNA全长序列进行分析。通过杜氏纤维素选择性培养基并结合测定以滤纸、脱脂棉和羧甲基纤维素钠为底物的纤维素酶活力,从中筛选出纤维素酶高产菌株5株。结果表明,在长期自然环境条件下,从云南云秃蝗肠道内分离的可培养22株细菌分别归属Bacillus sp.,Pseudomonas sp.,Nocardiopsis sp.,Kocuria sp.,Cellulosimicrobium sp.五大类菌群,其中文中所筛选出的5株具有纤维素降解功能的细菌中,4株为芽孢杆菌属,1株为假单胞属。纤维素酶活性测定结果表明,XN-80-5以滤纸和羧甲基纤维素钠为底物时纤维素酶活力最高,分别为167.49U/mL、573.57U/mL,而XN-80-1和XN-80-2均以脱脂棉为底物时纤维素酶活力最高,分别为32.85U/mL、33.76U/mL。22 cultivable bacteria were isolated from alimentary canal of Yunnanacris yunnaneus by screening techniques,and the phylogenetic diversities of them were analyzed by bacterial specific whole length 16S rDNA sequencing. Five high producing-cellulase strains were isolated in the cellulose selective medium according to their cellulose enzyme activity detected on the filter paper, absorbent cotton and CMC-Na. The results showed that the 22 cultivable bacteria isolated from alimentary canal of Yunnanacris yunnaneus were classified into five floras, including Bacillus sp., Pseudomonas sp., Nocardiopsis sp., Kocuria sp. and Cellulosimicrobium sp.. Among 5 cellulase strains of cultured bacteria, four were Bacillus sp. and one was Pseudomonas sp.. The cellulase activities analysis showed that XN-80-5 had the highest cellulase activity detected on filter paper and CMC-Na which were up to 167.49 U/mL and 573.57 U/mL,respectively. XN-80-1 and XN-80-2 had the highest cellulase activities detected on absorbent cotton which were up to 32.85 U/mL and 33.76U/mL, respectively.
