摘 要:目的了解胡桃楸提取物(JMME)治疗小鼠弓形虫感染的作用。方法建立弓形虫感染小鼠模型。将弓形虫感染的小鼠分成5组,分别以0.090、0.045和0.023mg/(kg.d)的JMME灌胃治疗,1次/d,疗程12d,同时设乙酰螺旋霉素(SM)治疗组和生理盐水(NS)对照组,观察各组小鼠的生存情况,弓形虫在小鼠体内的分布,计算胸腺指数和脾指数,MTT法测定淋巴细胞转化能力。结果JMME高、中、低剂量治疗均能推迟弓形虫感染小鼠的死亡时间,JMME低剂量治疗组和SM治疗组小鼠30d存活率分别为20.0%和15.0%,差异有统计学意义(P<0.05)。弓形虫在小鼠体内的分布以腹水居多,密度NS组高于各治疗组,肝、脾组织内弓形虫也以NS组居多。治疗后感染鼠体内的弓形虫未出现远端播散,除NS组外,其他各组脑组织内均未查到虫体。JMME高、中、低三剂量治疗组小鼠的胸腺指数分别为2.44±0.45、2.43±0.58和2.36±0.47,与NS组1.74±0.83比较差异有统计学意义(P<0.05);脾指数分别为4.35±0.42、4.41±0.25和4.43±0.13,与NS组3.17±0.06比较差异有统计学意义(P<0.05);淋巴细胞转化A570值分别为0.535±0.028、0.512±0.043和0.498±0.024,与NS组0.318±0.027比较差异有统计学意义(P<0.05)。结论JMME对小鼠弓形虫感染有一定的治疗作用。Objective To ascertain the action of an extract of Juglans mandshurica Maxim(JMME)in mice infected with Toxoplasma gondii.Methods A mouse model of T.gondii infection was established.The infected mice were divided into 5 groups and given three daily doses of JMME,i.e.0.090,0.045 and 0.023 mg/(kg·d)via intragastric administration for 12 days or were given acetylspiramycin(SM)or normal saline(NS)as a control.The survival of mice in each group was observed,the distribution of T.gondii in mice was determined,the thymus index and spleen index were calculated,and the lymphocyte transformation capacity of MTT was determined.Results High,medium,and low doses of JMME delayed the death of mice infected with T.gondii.The low-dose JMME group and the SM group had 30 d survival rates of 20.0% and 15.0%,and the difference was statistically significant(P0.05).T.gondii in mice was mostly found in ascites,and the NS group had more T.gondii than the treated groups(P0.05).The NS group had a large amount of T.gondii in liver and spleen tissues.After treatment of T.gondii infection,remote spread was not apparent in the mice.T.gondii was not found in the brain tissue of treated mice.Mice in groups receiving a high,medium,or low dose of JMME had a thymus index of 2.44 ± 0.45,2.43 ± 0.58,or 2.36 ± 0.47 while mice in the NS group had an index of 1.74 ± 0.83;the difference between the two was statistically significant(P0.05).Mice in groups receiving a high,medium,or low dose of JMME had a spleen index of 4.35 ± 0.42,4.41 ± 0.25,or 4.43 ± 0.13 while mice in the NS group had an index of 3.17 ± 0.06;the difference between the two was statistically significant(P0.05).Lymphocyte transformation A_570 values were 0.535±0.028,0.512±0.043,and 0.498±0.024 for the groups receiving a high,medium,or low dose of JMME but 0.318±0.027 for the NS group;the difference between the two was statistically significant(P0.05).Conclusion JMME has a therapeutic effect to an extent in mice infected with T.gondii.
