右归丸对雄激素致排卵障碍型不孕大鼠血清E_2、T和IGF-1的水平影响

摘　　要：目的研究右归丸对雄激素致排卵障碍型不孕大鼠血清中雌二醇(E2)、睾酮(T)和胰岛素样生长因子1(IGF-1)水平的影响,以进一步探讨右归丸治疗不孕症的作用机理。方法将43只9日龄SD雌性大鼠随机分为空白对照组、模型组、右归丸组、枸橼酸氯米芬组,建立雄激素致排卵障碍型不孕大鼠模型,第80日龄开始给药,5周后股动脉取血,用放免法检测血中E2和T水平,用酶联免疫法检测血清中IGF-1水平。并取出卵巢制作病理切片,HE染色,进行病理形态学观察。结果右归丸能够降低病理性囊性卵泡数,促进卵泡发育成熟及排出,并有黄体生成。右归丸能够增加血清中E2含量(P<0.05),降低血清中T的含量(P<0.05),右归丸有降低IGF-1趋势。结论右归丸有可能通过降低IGF-1和T、促进T转化为E2这一途径,从而达到促排卵和助孕安胎。Objective To study the impact of Yougui-Pill on androgen-induced anovulatory infer tility rats serum E2, T and IGF-1 levels, in order to further explore the mechanism of Yongui-Pill on infertility treatment. Methods 43 9-day-old SD female rats were randomly divided into blank control group, model group, Yougui-Pill group, clomiphene citrate group, and were induced to be anovulatory infertility by androgen. The medication started from 80-day-old And lasted for 5 weeks. Then the blood was taken through femoral artery and the level of serum E2 and T was measured by radioimmunoassay, IGF-I by enzyme-linked immunosorbent assay. Ovarian tissue was taken out for pathomorphology examination after HE dyeing. Results Yougui-Pill can reduce the number of pathological cystic follicles, promote follicular maturation and discharge and luteinize. Yougui-Pill can increase the E2 level in serum （ P 〈 0. 05 ）, decrease the T levels in serum （ P 〈 0. 05）. Yougui-Pill demonstrated the trendency of decreasing IGF-1. Conclusion Yougui-Pill can induce ovulation and assist tocolysis by reducing the IGF-1 and T level in se- rum and promoting the T changing into E2.
