口蹄疫A型病毒AF/72株免疫原性分析

摘　　要：用口蹄疫A型病毒AF/72株,经RT-PCR获得其VP1基因序列,并与GenBank中的其他5株A型口蹄疫病毒株比对,同源性均大于90%。将此病毒灭活后与弗氏佐剂联合制成灭活疫苗免疫豚鼠后分离血清,经液相阻断ELISA和微量细胞中和试验检测此血清的抗体效价,其均值分别为2.093和1.227。用微量细胞中和试验分别测定了AF/72参考血清对5株A型口蹄疫病毒的中和抗体效价,各试验重复3次,其平均值为2.156。分别计算AF/72对5株不同病毒株的r值,结果为0.79~0.92,均值为0.85。按照OIE标准,AF/72具有较强的免疫原性,抗原谱广,可作为制造口蹄疫A型疫苗的备选毒株。The VP1 gene sequence of foot-and-mouth disease virus（FMDV） type A strains AF/72 through RT-PCR was obtained.VP1 gene showed more than 90% DNA homology to other five type A strains of FMDV genes retrieved from NCBI GenBank.Serum was isolated from guinea pig injected with inactivated AF/72 of FMD type A virus and Freund′s adjuvant.By using ELISA and micro-neutralizing tests,the average antibody titers of this serum were 2.093 and 1.227,respectively.To determine antibody titers of AF/72 reference serum to five A-type FMDVs,three-repeat micro-neutralizing test was applied which showed the average value was 2.158.Calculated separately for five different strains of type A virus,the average r value of AF/72 was 0.85.According to OIE standard,AF/72 showed stronger immunogenicity,broader antigen spectrum,and could be used as an alternative stain for preparation of foot-and-mouth disease type A vaccine.
