三种梨果皮总RNA提取方法的比较研究

摘　　要：以西洋梨品种‘早红考密斯’、‘香红蜜’和白梨品种‘华酥’成熟果实为试材,采用Plantol总RNA提取试剂盒法、改良CTAB法和SDS法提取果皮中总RNA,并通过凝胶电泳、核酸蛋白分析仪和RT-PCR检测所提取总RNA样品的质量.结果表明,Plantol试剂盒法和改良CTAB法提取的RNA,经RT-PCR后得到的条带清晰,与目的片段大小一致,可以满足基因表达分析等试验的要求;而SDS法所提取的部分RNA,RT-PCR扩增不到目的片段,难以达到进一步试验的要求.The pericarp of pear fruit（cvs.＇Early Red Cornice＇＇Xianghongmi＇ and ＇Huasu＇） was used as material for extracting total RNA with Plantol total RNA extraction kit,improved CTAB method and SDS method.The quality of the total RNA was tested by nuclear acid/protein analyzer,gel electrophoresis and RT-PCR.The results showed that the RNA got from Plantol total RNA extraction kit and improved CTAB method had a better quality,with a distinct bands and the size as the same with target fragments,and could satisfy the requirement of molecular biology trial.However,the RNA got from SDS method did not reached the aim for next trials.
