TBTCl致PC-12细胞凋亡及Bax/Bcl-2表达的变化

摘　　要：目的研究氯化三丁基锡(TBTCl)致大鼠肾上腺嗜铬细胞瘤细胞(PC-12细胞)凋亡作用及Bax与Bcl-2 mRNA表达的变化。方法当PC-12细胞处于代数生长期时,分别加入不同浓度(0.000、0.013、0.025、0.050、0.100、0.200、0.400 mg/L)的TBTCl处理48 h,MTT试验检测细胞增殖;流式细胞仪检测细胞凋亡率;实时荧光定量PCR检测Bax和Bcl-2 mRNA的含量。结果随着TBTCl染毒浓度的增高,其对PC-12细胞增殖的抑制作用逐渐增强,当TBTCl浓度为0.400 mg/L时,其抑制率达到(68.25±3.83)%,IC50=0.15 mg/L,而细胞的凋亡率由对照组的(0.8±0.3)%上升到(25.4±1.5)%;TBTCl能上调Bax基因和下调Bcl-2基因的表达,0.400 mg/L TBTCl组Bax和Bcl-2分别是对照组的5.1±0.6倍和0.25±0.08倍。结论TBTCl能抑制PC-12细胞的生长,诱导细胞凋亡,其可能机制是上调Bax基因的表达和下调Bcl-2基因的表达。Objective To explore the effect of tributyltin chloride （TBTC1） on PC -12 cell of apoptosis, growth inhibiting, expression of Bax and Bcl - 2. Methods PC - 12 cell were incubated with various concentration of TBTC1 for 48 hours. The cell growth inhibition was measured by MTI＇, and apoptosis cell was observed by the flow cytometry analysis, and the expression of Bax and Bcl -2 were detected by real time PCR. Results TBTCl could result in descent of the cell proliferation （IC50 = 0. 128 mg/L） , induced the apoptosis in PC -12 cell. The rate of apoptosis of PC -12 cell reached to （ 25.4 ± 1.5 ） % at the TBTCl concentration of 0.400mg/L. The expression of Bax was significantly up - regulated and Bcl -2 expression was significantly down -regulated at higher concentration （0. 050,0. 100,0. 200,0. 400 mg/L）. Conclusion TBTC1 could induce the PC - 12 cell apoptosis, up - regulate the expression of Bax and down - regulate the expression of Bcl -2.
