摘 要:番茄叶片富含酚类、多糖、蛋白质等次生代谢物质,用普通方法提取番茄叶片总RNA时很难将其去除,试验比较研究了RNA plant Reagent、UNIQ-10柱总RNA抽提试剂盒和改进Trizol三种方法的提取效果。结果表明,改进的Trizol法能从番茄叶片中提取纯度较高的RNA,凝胶电泳显示条带清晰完整,分光光度计检测A260/A280比值为1.88。用该方法提取的番茄叶片总RNA可以用于逆转录、RT-PCR、Northern blot等分子生物学研究。Tomato leaves contain phenols, polysaccharide protein and other secondary metabolism substances. These components were difficult to eliminate by common methods when total RNA was extracted. RNA plant Reagent method, UNIQ-10 column kit method and the improved Trizol method were compared and researched. The results showed that the high purity RNA were obtained from the improved Trizol method from tomato leaves, gel electrophoresis showed clear and complete strips, and A260/A280 was 1.88. The RNA extracted by the improved Trizol method from tomato leaves could be used to reverse transcription, RT-PCR, and Northern blot.
