摘  要:以北极狐的生长激素基因(GH)作为候选基因,以大兴安岭图强林业局狐场提供的北极狐为试验群体,采用单链构象多态性(PCR-SSCP)和DNA测序的方法检测了GH基因单核苷酸多态性(SNPs),针对该群体的特点建立合适的统计分析模型,并进行了GH基因多态性与生长性状的相关分析。结果表明,在北极狐GH基因的外显子2、内含子2和外显子3上发现4个多态位点,分别为外显子2上的G81A突变、内含子2上的C186T和T239C突变,以及外显子3上的C583A突变;GH基因C583A多态性与公狐的体长性状显著相关(P<0.05)。利用以上点突变对北极狐的体重及皮张长度性状进行标记辅助选择研究,可达到快速选育出优质北极狐的目的。The effects of candidate gene growth hormone (GH) gene on growth traits in the current study. Single nucleotide polymorphisms (SNPs).of the GH gene were identified and genotyped by polymerase chain reaction followed by single strand conformation polymorphism (PCR-SSCP) analysis and DNA sequencing in a arctic fox population from the fox farm of Daxing'anling Tuqiang forest enterprise. Correlation analysis between GH polymorphisms and growth traits was carried out by using the appropriate mixed model. Results showed that four SNPs, G81A in the second exon, C186T and T239C in the second intron, C583A in the third exon, were found within arctic fox GH gene. The C583A polymorphisms of GH were associated with male fox body length (P〈0.05). In order to select big and quality arctic fox, the experiment could undertake marker assistant selection to body weight and the skin length of arctic fox by the polymorphic sites.
