摘  要:【目的】建立一高效液相色谱法同时测定人血清中咪哒唑仑(MZ)、阿普唑仑(AZ)、氯硝安定(CZ)药物浓度,为临床用药及治疗提供依据。【方法】采用反相高效液相色谱法,色谱柱为依利特Kromasil C18柱(250mm×4.6mm,5μm);柱温40℃;流动相为甲醇-水(65:35,v/v),流速1.0mL/min;波长220nm。血液样品采用10%高氯酸沉淀蛋白后,直接进样20μL,测定,采用内标法定量。【结果】MDZ、AZ、CZ在0.025~10.00μg/mL浓度范围内线性关系好,相关系数分别为r=0.9994,0.9993,0.9991,最低检测限均为5.0ng/mL,三种物质的平均方法回收率均大于92.0%,相对标准偏差(RSD)〈4.1%。【结论】本法测定MZ、AZ、CZ的血药浓度方法简便、快速准确,可用于临床的常规监测。[Objective] To develop a reverse phase high performance liquid chromatography(RP HPLC) for simultaneous determination of human serum concentration of midazolam(MZ), alprazolam(AZ) and clonazepam(CZ) and monitor the useage of these compounds. [Methods] Kromasil C18 column was used and the col umn temperature was 40 ℃. The mobile phase was methanol/water (65:35, V/V). The flow rate was 1.0 mL/min. The detection wave length was 220 nm. Twenty μL sample was injected after the plasma protein was precipitated with10% perchloric acid. Internal standard was used for quantitation. [Results] The linear relation of MZ, AZ and CZ was in the range of 0. 025-10.00 μg/mL. The minimum detection limit of MZ, AZ and CZ was 5.0 ng/mL. The average coefficient of recovery of three drugs was more than 92.0% and the relative standard deviation(RSD) was less than 4. 1%. [Conclusion]RP-HPLC method is a rapid, accurate and simple assay to measure MZ, AZ and CZ concentration in human serum and it can be applied routinely to monitor the concentration of these three drugs.
