摘  要:目的探讨鼻息肉组织中核因子кB(Nucler factorкB,NF-кB)的活化对IL-1a转录的影响及意义。方法取32例确诊为鼻息肉患者的息肉组织,以其中的14例患者的鼻中隔黏膜作对照,用凝胶迁移实验(Electrophoreticmobility shift assay,EMSA)检测NF-кB的DNA结合活性,用RT-PCR检测IL-1a mRNA的表达,对NF-кB的DNA结合活性与IL-1a的转录水平行Pearson相关性分析。结果鼻息肉组织中NF-кB的DNA结合活性升高,明显高于对照组(p<0.01);鼻息肉组织中IL-1a mRNA的表达明显高于对照组(P<0.01);Pearson相关性分析提示,鼻息肉组织中NF-κB的DNA结合活性与IL-1a的转录呈显著正相关(r=0.777,P<0.01)。结论IL-1a是鼻息肉中NF-кB激活后所分泌的重要的细胞因子,NF-кB和IL-1a引起的局部微环境的改变可能是鼻息肉发生的重要内在因素。Objective To investigate the effects of NF-κB activation on transcription of IL-1a in the tissues of nasal polyps. Methods The specimens from 32 nasal polyps and 14 nasal septum mucous membranes were studied by electrophoretic mobility shift assay(EMSA)for the activation of NF-κB and by reverse-transcription polymerase chain reaction (RT-PCR) for the expression of IL-1a mRNA. The correlations between the activation of NF-κB and the transcription of IL-1a mRNA were evaluated by Pearson correlation analysis. Results The activation of NF-κB DNA were significantly increased in the tissues of nasal polyps compared that of nasal septum mucous membranes (P〈0.01). The expression of IL-1a mRNA were significantly increased in nasal polyps than that in controls (P〈0.01). The Pearson correlation analysis demonstrated that there were positive relationship between the activation of NF-κB DNA and the transcription of IL-1a (r=0. 777,P〈0.01). Conclusion These results suggest that IL-1a regulated by NF-κB may be a key cytokine in nasal polyps. It should he possible that the changes of microenvironment caused by NF-κB and IL-1a may play an important role in the mechanisms for the pathogenesis of nasal polyps.
