摘  要:目的研究猫爪草多糖(RTP)对小鼠急性化学性肝损伤的保护作用及其机制。方法将小鼠随机分为正常组、模型组、联苯双酯阳性对照组、RTP低、中、高剂量组(RTP剂量分别为100、200、400mg·kg-1.d-1),四氯化碳(CCl4)调和油溶液腹腔注射建立小鼠急性肝损伤模型,测定肝体指数、血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、肝组织匀浆的总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)活力、丙二醛(MDA)含量,观察小鼠肝组织病理改变。结果 RTP组均能降低CCl4所致小鼠急性肝损伤肝体指数及血清ALT、AST活性,提高肝组织T-AOC、SOD活力,降低MDA含量,且可以不同程度地改善肝组织损伤。结论 RTP对CCl4所致小鼠急性化学性肝损伤有保护作用,其机制可能与RTP抗氧化作用有关。Objective To study the protective effect of polysaccharide of radix ranunculi ternati(RTP) on CC14-induced acute liver injury in mice. Methods Kunming mice were randomly divided into control group, model group, bifendate group and low,medium,high dosage of RTP groups. Three RTP subgroups were given RTP for 10 days by the body weight dosage 100,200,400 mg·kg-1· d-1 respectively. Mice in the control group were injected with oil solution through abdominal cavity,and mice in the other groups were given CCl4 mixed with oil solution 2 hours after the last administration. The activities of ALT and AST,the content of T-AOC,SOD, MDA and liver in dex,as well as the pathological changes of the liver were determined in all groups. Results RTP can reduce the liver index of mice,serum activities of ALT,AST and hepatic MDA. It can also raise the liver T-AOC,enhance the activities of SOD in liver tissue and improve the liver histology. The best dosage of RTP was in the fixed scope. Conclusion RTP can protect the acute liver injury induced by CCl4. The mechanism may be related to free radical scavenge and lipid peroxidation.
