摘 要:目的:研究"主客交"理论指导下透邪通络法抗肝纤维化的机制及效果。方法:选用SD雄性大鼠,40%CCl4腹腔注射造模,正常组和模型组予生理盐水灌胃,对照组予秋水仙碱灌胃,透邪通络组予加减三甲散灌胃,8周后处死大鼠,显微镜下观察肝组织形态,免疫组化法检测MMP-1、TIMP-1的表达,用图像分析软件和统计软件处理结果。结果:与模型组相比,透邪通络组升高MMP-1表达与降低TIMP-1表达均有显著差异(P<0.01),对照组升高MMP-1表达有差异(P<0.05)、降低TIMP-1表达有显著差异(P<0.01);与对照组比较,透邪通络组提高MMP-1表达、降低TIMP-1表达均有差异(P<0.05)。结论:透邪通络法能明显改善HF大鼠的病理,具有显著的抗肝纤维化,其机制可能与提高MMP-1表达、降低TIMP-1表达而调控ECM的代谢有关。Objective:To study the mechanism and effect of Touxietongluofa on hepatic fibrosis under the guidance of Zhukejiao theory. Methods:Male SD rats except the rats in the normal group, others were inflicted with liver fibrosis by intraperitoneal injection of 40% carbon tetrachloride,and were given medication at the same time. Rats were given normal saline in the normal group and the model group, colchicine in the positive group ,Jiajiansanjiasan extraetum in the Touxietongluo group. After eight weeks, all the rats were execu- ted, and the hepatic tissues were kept for pathologic examination to observe hepatic fibrosis and immunohistoehemical to detect MMP-1 and TIMP-1 of hepatic tissue. The experimental results were processed by image system and statistical software. Results : Compared with the model group, MMP-I expression increased obviously and TIMP-1 expression decreased obviously in the hepatic tissues of rats in the Touxietongluo group( P 〈 0.01 ) ,while MMP-1 expression increased ( P 〈 0.05 ) and TIMP-1 expression decreased obviously ( P 〈 0. 01 ) in rat liver of the positive group. Compared with the positive group, MMP-1 expression increased and TIMP-1 expression decreased in the hepatic tissues of rats in the Touxietongluo group( P 〈 0.05 ). Conclusions :Touxietongluo can obviously relieve the pathological damage in hepatic tissues of hepatofibrosis rats, and thus has a good curative effect of anti- hepatic fibrosis. Its therapeutic mechanism mainly maybe adjust the metabolism of extra-cellular matrix by increasing MMP-1 expression and decreasing TIMP-1 expression.
