玻璃化冷冻人成熟卵母细胞的临床应用

摘　　要：目的探讨玻璃化冷冻在人成熟卵母细胞冻存中的临床应用效果。方法运用cryoleaf进行人类成熟卵母细胞玻璃化冷冻保存,冻融后成功复苏的卵母细胞行胞质内单精子注射-胚胎移植(ICSI-ET),观察复苏后卵母细胞受精、卵裂以及生化妊娠、临床妊娠情况。结果15例共81枚成熟卵母细胞冻融后进行卵胞浆内单精子注射或者体外受精-胚胎移植(IVF-ET),有69枚卵母细胞成功复苏,52枚卵母细胞受精,39枚卵母细胞卵裂,移植后4例患者获得临床妊娠。结论玻璃化冷冻人成熟卵母细胞方便、快捷、成本低下、冷冻损伤小,有极大的临床应用价值。Objective To explore the clinical application value of oocytes cryopreservation.Methods Human mature oocytes were cryopreserved using cryoleaf.After thawing,all survived occytes at metaphaseⅡ（MⅡ） were performed for intracytoplasmic sperm injection （ICSI）.Then,all fertilized occytes were picked up for a further culture.In D2 or D3,high quality embryos were selected for embyo transfer.Furthermore,the fertilization,cleavage,developmental capacity of oocytes and the result of clinical pregnancy were observed and detail recorded.Results After thawing,69 of 81 cryopreseved occytes from 15 patients were survived.All these 69 survived oocytes were performed for ICSI.Subsequently,52 oocytes were found for fertilization,39 were found for cleavage,4 patients got clinical pregnancy.Conclusion Vitrification of human mature oocytes is convenient,fast,cost low,and small freeze damage,it has valuable in clinic.
