维生素D受体基因启动子区CDX2多态性与儿童氟斑牙的关系

摘  要:目的了解维生素D受体(VDR)基因启动子区CDX2结合位点多态性与儿童氟斑牙易感性的关系。方法Dean法检查8~12岁凤台县关店乡儿童氟斑牙患病情况,收集氟斑牙患者102例及正常对照者107例,采集外周静脉血提取DNA,采用等位基因特异多重聚合酶链反应(ASM-PCR)测定两组儿童VDR基因CDX2结合位点多态性。结果VDR基因CDX2结合位点多态性分布频率为:氟斑牙患者GG占48.0%、AG占25.5%、AA占26.5%;正常对照组GG占55.1%、AG占28.0%、AA占16.9%。两组基因型分布比较差异无统计学意义(P>0.05)。结论VDR基因CDX2结合位点多态性与所调查儿童的氟斑牙发生无明显关系。Objective To investigate the association of polymorphisms of CDX2 binding site in vitamin D receptor gene(VDR) with the susceptibility to children′s dental fluorosis.Methods A case-control study was conducted among children between 8 and 12 years old with (n=102) and without(n=107) dental fluorosis using Dean′s method in GuanDian countyside of FengTai county.DNA was extracted from blood samples of these children.VDR gene CDX2 polymorphisms were analyzed using allele-specific multiplex PCR(ASM-PCR).Results The frequency distribution of CDX2 genotype was GG 48.0%,AG 25.5%,AA 26.5% in children with fluorosis,and GG 55.1%,AG 28.0%,AA 16.9% in children without fluorosis respectively.There were no significant differences in the two groups(P〈0.05).Conclusion There are no correlation between VDR gene CDX2 polymorphisms and the children′s dental fluorosis in this study.
