摘  要:采用ISSR分子标记对裕丹参4种变异类型基因组DNA进行PCR扩增,利用SPSS11.5软件进行类型间亲缘关系分析,构建遗传树状图.结果表明,从50条ISSR引物中筛选出8条具有鉴别意义的多态性引物,在裕丹参4种变异类型中共得到65条带,47条为多态性条带,多态性位点72.3%,聚类分析将裕丹参4种类型分为2个类群,表明裕丹参4种变异类型遗传分化明显.ISSR标记可以作为裕丹参4种变异类型鉴定的有效分子标记.PCR amplification was performed with the genomic DNA of four variance-types of Salvia miltiorrhiza by ISSR molecular markers. The genetic relationships were analyzed by using analysis software SPSS11.5, and the DNA molecular dendrogram was established. The results showed that eight polymorphic primers for identification were chosen from 50 ISSR primers. A total of 65 bands were amplified from four variance-types of S. miltiorrhiza using the eight screened primers, including 47 polymorphie bands taking up about 72.3%. A systematic cluster analysis conducted shows that the four variance-types of S. miltiorrhiza can be divided into two groups, which suggests that there is abundant genetic diversity among the four variance-types of S. miltiorrhiza, and ISSR markers can be effectively used to identify the four variance-types of S. miltiorrhiza.
