摘 要:[目的]探索胶原-透明质酸钠-纤维蛋白胶多孔支架复合去抗原松质骨的骨软骨一体化材料的制备方法及其特征。[方法]将胶原、透明质酸钠及纤维蛋白胶混匀,利用纤维蛋白胶的粘合性与去抗原牛松质骨复合,成为骨软骨一体化的支架材料。通过大体观察和扫描电镜观察了解材料的孔径和交通情况;分离、增殖仔兔骨髓基质细胞(BMSCs),取第3代BMSCs接种至支架材料复合培养,1、3、5、7和10 d后,行MTT检测并绘制细胞生长曲线;取第7 d标本,行扫描电镜观察。[结果]经交联和复合的一体化骨软骨支架材料具有良好的三维多孔结构,骨相孔径约300~500μm,软骨相孔径约80~120μm,两部分连接紧密;电镜观察见骨相及软骨相均有细胞黏附,细胞周围有基质存在;MTT检测显示,各时间点的OD值在实验组与对照组之间无统计学差异(P〉0.05)。[结论]胶原-透明质酸钠-纤维蛋白胶多孔支架复合去抗原牛松质骨的一体化材料具有良好的空间结构和生物相容性,可用于修复骨软骨联合缺损的组织工程支架材料的研究。[Objective]To prepare and investigate a scaffold made of collagenⅠ-sodium hyaluronate-fibrin glue tri-copolymer bonding with antigen-extracted bovine cancellous bone in vitro. [Methods]The osteochondral scaffold was constructed of collagenⅠ-sodium hyaluronate-fibrin glue tri-copolymer scaffold and antigen-extracted bovine cancellous bone with adhesion of the two parts by fibrin glue.Pore size and internal pores of the osteochondral scaffold were observed by gross and scanning electron microscopy(SEM).Rabbit bone marrow stromal cells(BMSCs) were isolated and amplified before they were seeded into the scaffold at the third passage.The proliferation of the cells on the scaffolds was examined using MTT method and the growth curve was drawn.The scaffolds with cells were dehydrated and observed by SEM after 7 days.[Results]The osteochondral scaffold after crosslinking and adhesion had the porous structure.The bone scaffold was bonded to the cartilage scaffold solidly.The examination under SEM indicated that the BMSCs adhered and spread well on the scaffold,and the extracellular matrices were also observed around the BMSCs.MTT method revealed that there was no significant difference between the experimental group and the control group at each time point(P0.05).[Conclusion]The osteochondral scaffold of the collagenⅠ-sodium hyaluronate-fbrin glue tri-copolymer scaffold bonding with antigen-extracted bovine cancellous bone has an appropriate structure and a good biocompatibility,which makes it a useful scaffold in the osteochondral tissue engineering.
