摘　　要：目的探讨肿瘤坏死因子（tumornecrosisfactor，TNF）对体外培养小梁细胞整合素（integrin）表达的影响及意义。方法体外培养的牛眼小梁细胞分别以TNF-α浓度0．0mg/ml、20ng／ml、40ng／ml、60ng／ml孵育36h，免疫组化法行整合素-β1染色后用倒置显微镜观测，RT—PCR半定量检测整合素α-5表达量，利用凝胶图像分析系统对实验结果图像分析。结果实验组阳性反应图像积分光密度值高于对照组，且随TNF—α浓度增高呈线性递增关系。RT—PCR法integrinα-5／β-actin光密度比值与对照组差异具有统计学意义。结论在体外培养条件下TNF—α可引起牛眼小梁细胞整合素合成增加，整合素或许在选择性激光小梁成形术降低眼压方面发挥了一定作用。Objective To investigatetheeffect Of tumomecrosisfactor-α （TNF-α）On mtegnn expression In cultured bovine trabecular meshwork cells （TM cells）. Methods Cultured bovine TM cells were incubated with TNF-α （divided into0.0ng·ml^-1,20ng ·ml^-1,40ng ·ml^-1,60ng ·ml^-1）for 36h,stained with integrin-β 1by tmnlunolllstochemistry and observed with inverted microscope. Measuring Integrm-α 5 byseti-quantitativeRT-PCR, theexperimentalresultswereanalyzedwith computerized unage analys-istechniques. Results Integrin in experimental Rroup Has stronger posmve reaction than control group,it presents linear dependence with the TNF-increasing by statistical analysis. The difference value Of integrinα-5 / β -aetin between treated groups and that of control group was statistically significant. Conclusion TNF-α Can caused an increase in levels or integrill in bovine, Fmeells in vitro, integrin may play al 1 important role in developing the mechanism Of decreasing intraocular pressure in select trabeeuloplasty.
