摘 要:目的 观察骨髓基质干细胞(MSCs)/煅烧骨/Ⅰ型胶原复合物修复颅骨缺损的能力.方法 18只成年Wistar大鼠颅骨分别制作直径8 mm圆形全层骨缺损.其中6只在全身麻醉及无菌条件下取其左侧股骨,诱导培养的骨髓基质干细胞与煅烧骨/Ⅰ型胶原复合制成复合物修复颅骨缺损,为实验组;6只以煅烧骨/Ⅰ型胶原修复为对照组;6只不修复为空白组.于植入复合物后4周和8周两个时间点处死大鼠,以组织学方法 观察骨修复情况.结果 4周时实验组的植入物与颅骨缺损创缘呈骨性连接,8周时实验组植人物与创缘呈骨性连接,小梁骨增多,小梁间有较多的骨髓组织形成.对照组植入物与创缘以纤维连接为主,极少部分边缘可见骨性连接.空白组颅骨缺损面积未见明显缩小.结论 MSCs和胶原化的煅烧骨有非常好的生物相容性,复合后植入体内能够成骨,可用于颅骨缺损的修复.Objective To investigate the effect of compound of ceramic bovine bone(CBB)/collagen I /MSCs on repairing cranial defects. Methods A full-thickness cranial defects with 8 mm in diameter were created in 18 adult Wistar Rats. MSCs from 6 Wister Rats were cultured, expanded and induced into osteoblasts in vitro, and mixed with CBB/collagen I . Cranial defects were repaired with CBB/collagen I mixed with MSCs in 6 rats ( the experimental group), repaired with CBB/eollagen alone in 6 rats ( the control group), 6 rats were without repair (the blank group). The animals were sacrificed at 4 weeks and 8 weeks and histological observation of new bone regeneration in the defects was performed. Results In the experimental group, histological analysis showed that osseous connection between the implanted compound and the edge of the defects at 4 weeks, increased trabecular bone and much myeloid tissue among trabecula at 8 weeks, fiber connection with slight osseous connection appeared in the control group, no obvious changes happened in the blank group. Conclusion The compound of MSCs with CBB/eollagen is a good material for repairing the skull defects with such advantages as biocompatibility and osteogenesis.
