摘 要:目的 观察苦杏仁甙对博莱霉素诱导的肺纤维化大鼠血清生物标志物的影响.方法 将72只Wistar大鼠随机分为对照组、博莱霉素组和苦杏仁甙组,每组24只.分别观察7、14、28 d.博莱霉素组和苦杏仁甙组气管内一次性注入博莱霉素5 mg/kg,对照组气管内注入等体积生理盐水,同时苦杏仁甙组大鼠每天腹腔注射苦杏仁苷15 mg/kg.取肺组织制作石蜡切片,天狼猩红染色法结合偏振光显微镜观察肺组织I、Ⅲ型胶原的表达.表面增强激光解吸附离子化飞行时间质谱(SELDI-TOF MS)结合弱阳离子蛋白芯片检测各组大鼠混合血清差异蛋白的表达.结果 肺组织石蜡切片天狼猩红染色显示肺纤维化动物模犁肺组织存在持续纤维化反应.模型建立7、14、28 d后,3组均检测到4种差异表达的蛋白,其质荷比分别为3530.7,7043.5,8332.6和9068.0.与对照组比较,博莱霉素模型组7和28 d时质荷比为3530.7的血清蛋白标志物峰强度比值在〉2,在7、14、28 d时质荷比为7043.5,8332.6和9068.0的血清蛋白标志物峰强度比值均〉2,有明显差异.与博莱霉素组比较,苦杏仁甙组质荷比为7043.5(7 d),质荷比为8332.6(28 d)和9068.0(14 d)的血清蛋白标志物峰强度比值均〉2,有明显差异.结论 苦杏仁甙可抑制博莱霉素诱导的大鼠血清蛋白标志物峰强度的增加.Objective To evaluate effect of amygdlin on expression of four biomarkers in the animal model of pulmonary fibrosis induced by bleomycin.Methods Rats were given one dose (5 mg/kg)of bleomyem in bleomycin-treated groups,amygdlin-treated groups and saline in controls by intratrachealinstil.lation exposed surglcally,The amygdlin-treated groups rats were treated with intraperitoneal injeetion of amygdlin(15 mg·kg^(-1)·day^(-1)).The rats were sacrificed 7,14 and 28 days after bleomycin administration.Polarized light microscopy and Image-Pro Plus detected I and Ⅲcollagen expressed in Paraffin-embedded lung sections stained with Sirius red.Surface-enhanced laser desorption-ionization time-of-flight mass spectrometry (SELDI-TOF MS)with weak cationic proteinchip(CMiO)detected differentially expressed proteins in the pooled serum samples of all groups.Results Consistent fibrotic responses were found in all bleomycin and amygdlin-tread groups.On the 7th,14th and 28th day after bleomycin or saline instillation.four differentially expressed proteins were detected in the pooled serum of all groups rats,consisting of 4 proteins with mass/charge ratio of 3530.7,7043.5,8332.6 and 9068.0,respectively.Compared with control groups,pmtein peaks intensity ratio with mass/charge ratio of 3530.7 on 7,28 d and 7043.5,8332.6 and 9068.0 on 7.14 and 28 d was〉2 in bleomycin-treated groups.Compared with amygdlin-treated groups,protein peaks intensitv with mass/charge ratio of 3530.7 at 7,14,28 d had no change almost,but protein peaks intensitv ratio with mass/charge ratio of 7043.5 at 7 d,8332.6 on 28 d and 9068.0 on 14 d was〉2 in bleomycin-tread grouDs.All the four protein peaks intensity had no change almost at other point.Conclusion Amygdlin may reduce the bleom-ycin-induced increase of differentially expressed protein peak intensities in rat serum.
