白细胞介素32在博来霉素诱导的小鼠肺纤维化组织中的表达及意义

摘　　要：目的探讨白细胞介素32(IL-32)在博来霉素(BLM)诱导的小鼠肺纤维化过程中各期的表达和意义。方法将72只雄性小鼠随机分为三组,每组24只。分别给予0.9%氯化钠注射液0.04ml(C组)和等量的BLM(5mg/kg)(B、A组),一次性气管内灌注。A组在BLM灌注后每天给予地塞米松5mg/kg腹腔注射。然后各组分别于第3、7、14和28天各处死6只小鼠,取肺组织,做病理切片、免疫组织化学染色,应用Westernblot法测定IL-32表达。结果B组肺组织病理切片按时间顺序呈现由肺泡炎至纤维化动态改变。B组肺组织中IL-32蛋白表达水平高于C组(P<0.01)和A组。结论IL-32在肺纤维化形成过程中高表达,可能在肺纤维化形成的早中期发挥重要作用。Objective To study the expressions and significance of interleukin-32（IL-32） in lung tissues of mice with pulmonary fibrosis（PF） induced by bleomycin.Methods Seventy-two male mice were randomly divided into three groups with 24 rats each.The mice were given intratracheal instillation of bleomycin in groups of A and B.Normal saline was used instead of bleomycin in control group C.The mice of group A were injected abdominally with dexamethasone 5mg/kg every day.On the 3^rd,7^th,14^th and 28th day after administration,6 mice of each group were sacrificed and the lung tissues were harvested.Immunohistochemical staining and pathological examing of lung tissues were performed and the IL-32 expression of lung tissues was detected with Western blotting.ResultsDynamic changes from alveolitis to pulmonary fibrosis could be observed in group B.The expression of IL-32 in lung tissues was higher in group B than that in groups of C and A（P〈0.01）.ConclusionThe expression of IL-32 protein is highly expressed,which may be related to bleomycin-induced pulmonary fibrosis in the early and middle stages.
