阿苯达唑治疗沙鼠泡球蚴感染的实验观察

摘  要:为重新评价阿苯达唑对泡球蚴的杀虫作用,将34只沙鼠腹腔内移植感染泡球蚴后,随机分成3个治疗组和3个对照组,分别于感染后30d、50d和7d用含阿苯达唑005%的药饵喂饲(剂量50mg/kg/d),连续治疗6、10和7月,停药3月后解剖。结果显示3个治疗组的泡球蚴组织较对照组明显缩小,重量下降,减重率分别为867%、933%和957%,其中有3/6、3/6和4/5的沙鼠中泡球蚴重量明显低于感染时的01g。同时发现有13只治疗组沙鼠泡球蚴涂片镜检无活的原头节。病理学检查显示泡球蚴组织结构受到严重破坏,囊泡塌陷,生发膜坏死脱落,原头节崩解与钙化,形成异物肉芽肿和纤维化。实验表明,阿苯达唑不仅抑制泡球蚴的生长。Thirty four healthy gerbils infected with larval E.multilocularis tissue by intraperitoneal transplantation were divided into three treated and three control groups randomly. Thirty, fifty and seven days after infection these three group of gerbils were treated with 0.05% albendazolemedicated feeds (the equivalent to dosage of 50mg/kg/d) for 6, 10 and 7 months respectively. All gerbils were necropsied after 3 months of drugfree interval. The results showed that the larval weights of three treated groups of gerbils were markedly reduced in comparsion with those of control groups, the mean reduction rates were 86.7%,93.3% and 95.7% respectively. 3/6, 3/6 and 4/5 in three treated group gerbils, the larval tissue weights were below 100mg (inoculated transplant's weight). Viable protoscoleces were not found in 13 out of 17 treated gerbils in the minced larval tissue under microscopy examination. Histopathological examination of larval tissue in treated gerbils indicated that metacestode showed marked degeneration and necrosis with loss of structural integrity, the protoscoleces were mostly calcified, embeded in thick fibrous tissue. The experiment suggests that continuous longterm treatment with albendazole is parasiticidal to E.multilocularis, particularly in gerbils of early stage of infection.
