摘 要:目的观察大黄虫丸对大鼠动脉粥样硬化(AS)斑块及CD40表达的影响。方法将40只SD大鼠随机分为5组,即空白对照组、模型组、大黄虫丸高剂量组、大黄虫丸低剂量组及血脂康组,每组8只。采用经典喂养法复制动脉粥样硬化大鼠模型。模型复制成功后,空白对照组及模型组予蒸馏水按容积10mL/kg灌胃;大黄虫丸高、低剂量组予大黄虫丸溶液按生药1.4、0.7g/kg,容积10mL/kg灌胃;血脂康组混悬液按0.3mg/kg,容积10mL/kg灌胃。每日1次,连续8周。8周断头处死,采用酶比色法测定血清甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)含量;免疫组织化学法检测CD40表达;光镜下观察各组大鼠病理学形态变化。结果模型组与空白对照组比较,TG、TC、LDL-C含量增高,HDL-C含量降低(P<0.05),说明造模成功。与模型组比较,大黄虫丸高剂量组TG、TC、LDL-C降低,HDL-C升高(P<0.05);大黄虫丸低剂量组TG、LDL-C降低(P<0.05)。与血脂康组比较,大黄虫丸高、低剂量组CD40表达弱阳性(±)或阳性(+);大黄虫丸高、低剂量组与模型组比较,动脉粥样硬化斑块亦明显改善。结论大黄庶虫虫丸具有抗动脉粥样硬化作用,其机制可能与降血脂和下调CD40表达有关。Objective To investigate Dahuang Zhechong pill on the formation of atherosclerotic plaque and expression of CD40. Methods The research used rats to reproduce the model by feeding, 40 SD rats were divided into 5 groups randomly: control group, model group, Xuezhikang group, low dose group and high dose group. The rats in the control group were fed with normal forage and water. Others were fed with forage of high cholesterol and lipid. After atherosclerosis was established, the rats in the low dose group were fed with 0.7 g Dahuang Zhechong pill ( 10 mL) per lkg body weight and in the high dose group with 1.4 g. The rats in Xuezhikang group received intragastric 0.3mg Xuezhikang per 1 kg body weight. All rats in five groups were fed 8 weeks continuously. TC, TG, HDL - C and LDL - C in serum were measured by colourimetry. The expression of CD40 was measured by immunohistochemical method. Morphologic changes in rats were observed with optical microscope and electron microscope. Results There were significant differences between high dose group and model group on TG, TC, HDL - C and LDL - C ( P 〈 0.05 ). TG, TC and LDL - C were decreased, and HDL - C was increased. TG and LDL - C in low dose group was decreased in comparison with that in model group ( P 〈 0.05 ). The expression of CD40 in high dose group was weakly positive. The pathological morphous in low dose and high dose group were obviously changed.Conclusion Dahuang Zhechong pill has anti -arthros clerosis, its mechanism maybe related to decrease blood lipidand down regulate the expression of CD40.
