摘 要:目的了解Thrombopoietin(TPO)对人原巨核细胞白血病细胞株MO7e细胞的影响,间接了解TPO使用剂量和时机与血小板动力学的关系。方法MTT法测定MO7e细胞的计数,得出TPO浓度—MO7e细胞增值曲线,倒置显微镜下观察MO7e细胞的计数和形态。结果TPO可提高MO7e细胞增值,当TPO浓度为256ng/ml时细胞计数达到最高,加入TPO48h后MO7e细胞计数达到最高。结论TPO可促进MO7e细胞增值,TPO最佳的用药剂量和用药时机是256ng/ml和48h。Objective To observe the effect of thrombopoietin(TPO) on MO7e cells and the relation between its dose,use of time and dynamics of platelets.Methods MO7e cells were calculated by MTT for the TPO concentration-MO7e cell growth curve.The number and morphology of MO7e cells were observed under an inverted microscope.Results TPO could stimulate the growth of MO7e cells.The number of MO7e cells reached its peak when TPO was used at the concentration of 256ng/ml or 48h after TPO was used.Conclusion TPO can stimulate the growth of MO7e cells when it is used at the concentration of 256ng/ml and at the time of 48h.
