苦豆子总碱对急性期溃疡性结肠炎小鼠血清IL-1β,IL-4的影响

摘　　要：目的:观察苦豆子总碱(total alkaloid of Sophora alopecuroides,TASA)对葡聚糖硫酸钠(dextran sulfate sodium,DSS)致急性期溃疡性结肠炎(UC)小鼠的作用及对血清IL-1β,IL-4水平的影响。方法:C57BL/C小鼠随机分为6组:对照组、模型组、总碱高、中、低剂量组、柳氮磺吡啶组。5% DSS水溶液自由饮用8d以产生小鼠急性期UC模型同时灌服相应药物。观察小鼠一般活动状态及大便性状、潜血便血情况,结肠大体形态学及组织病理学改变,进行疾病活动指数评分(disease activity index,DAI),酶联免疫吸附法(ELISA)法检测小鼠血清IL-1β,IL-4的水平。结果:苦豆子总碱能显著改善DSS致结肠炎小鼠的一般状态,减轻其结肠组织病理学损伤。模型组小鼠结肠黏膜DAI比正常组明显增高(P<0.01)。而各给药组较模型组DAI均有不同程度的降低(P<0.01)。IL-1β与模型组相比均有不同程度的降低(P<0.01),而IL-4在数值上与模型组相比有所升高,但不具有统计学意义。结论:TASA对UC小鼠急性期损伤的大肠黏膜有明显的修复作用,可能通过抑制肠道免疫反应,减少促炎因子的过度表达,调节促炎因子与抗炎因子间的平衡而发挥其抗炎作用。Objective： To investigate the effects of total alkaloid of Sophora alopecuroides （TASA） on expression of serum IL- 1β and IL-4 in mice with DSS-induced acute ulcerative colitis. Method： C57BL/C mice were randomly divided into six groups： normal group, model group, SASP group, low, middle and high dosage TASA group. Acute ulcerative colitis was induced by administrating 5% dextran sulfate sodium（DSS）solution as drinking water freely for 8 days. Mice were given drug by stomach at the same time and sacrificed after 8 days. The general morphological and histopathological changes were observed during the experiments. Daily disease activity index （DAI） was assessed. The serum levels of IL-1β and IL-4 were detected by ELISA. Result： In UC mice, the expression of serum IL-1β was significantly increased whereas IL-4 was decreased when compared with the normal group （P 〈 0. 01 ）. In all treatment groups, TASA could reduce the expression of IL-1β （P 〈 0. 01 ）, increase the level of IL-4 and alleviate the symptom of UC and decrease the DAI. Conclusion： TASA can effectively attenuate DSS-induced acute ulcerative colitis through the decreasing the over-expression of pro-inflammatory cytokine levels.
