补肾复方对压力负荷增加大鼠血浆内皮素与降钙素基因相关肽水平的影响

摘  要:目的:观察补肾复方对压力负荷增加大鼠血浆内皮素和降钙素基因相关肽水平的影响。方法:60只雄性SD大鼠随机分为5组:假手术组、模型组、卡托普利组、补肾复方大剂量组、补肾复方小剂量组,每组12只。观察各组左室重量指数、左室心肌病理形态HE染色,采用放射免疫分析法检测血浆内皮素和降钙素基因相关肽的浓度。结果:①模型组左室重量指数、血浆内皮素(ET)浓度较假手术组明显增高(P<0.01),而降钙素基因相关肽(CGRP)下降,治疗后左室重量指数、ET浓度较假手术组下降明显(P<0.01),CGRP增高明显(P<0.05)。ET/CGRP比值在假手术组小于1,在模型组接近1,在卡托普利组、补肾复方大剂量组和小剂量组明显小于1,且这种改变趋势与左室重量指数呈正相关。结论:补肾复方通过降低ET和增高CGRP,能够对压力负荷增加致大鼠左室肥厚心肌细胞起到保护作用,其疗效与卡托普利相当。Objection:To observe the effects of Bushen prescription on plasma endothelin and calcitonin gene-related peptide(CGRP) in rats with pressure overload. Methods:Sixty male SD mice were randomly divided into 5 groups: sham group,model group,eaptopril group,high-dose Bushen prescription group and low-dose Bushen prescription group,with 12 mice in each group. Left ventricular mass index(LVMI),left ventricular myocardium HE staining, plasma endothelin(ET) and calcitonin gene-related peptide(CGRP) were measured in each group. Results:Compared with sham group,the LVMI,ET of model group increased but CGRP decreased. After treatment,ET decreased(P〈 0.05) but CGRP increased markedly(P〈0.05). Plasma ET/CGRP ratio was less than 1 in sham group obviously,that close to 1 in model group and obviously less than 1 in captopril group,high-dose and low-dose Bushen prescription. This tendency of changes positively related to LVH. Conclusion:Bushen prescription can protect myocardial cells of left ventricular hypertrophy caused by pressure overload through decreasing ET and increasing CGRP,and its effect is similar to captopril.
