新伐他仃对单肾切除糖尿病老龄鼠肾脏波形蛋白表达的影响

摘　　要：目的:观察辛伐他仃对单肾切除糖尿病老龄鼠血生化指标、波形蛋白表达,探讨新伐他仃的肾脏保护作用。方法:32只15月龄清洁级SD大鼠随机分为正常对照,肾切对照,糖尿病组,新伐他仃组。给予相应治疗后12周测定大鼠生化指标,对肾组织HE、PASM染色,镜下观察组织,免疫组化检测波形蛋白表达。结果:糖尿病组生化指标较对照组显著升高﹙P<0.05﹚;肾小球系膜增厚,足突融合,基底膜增厚﹙P<0.05﹚;肾小管扩张,上皮细胞水肿,损伤指数显著升高﹙P<0.05﹚;治疗组尿素氮、肌酐等较糖尿病组显著下降﹙P<0.01);治疗组甘油三酯、胆固醇与糖尿病组相比无明显变化﹙P﹥0.05﹚;治疗组与糖尿病组相比波形蛋白表达显著下降﹙P<0.01﹚。结论:治疗后肾脏病理改善,免疫组化发现新伐他仃减少波形蛋白异常表达。Objective：To explore the protective effect of simvastatin on the kidney of uninephrectomlzed STZ induced diabetic agng rat. Method：Thirty two fifteen -month -old SD rats were randomly assigned to normal control group, unlnephrectomlzed control group, simvastatin treatment group and Diabetic group After 12 weeks, biomedical markers were determined, the renal pathology changes were observed under light and electron; expression of vimentin were detected by immunohistechemistry. Results：biomedical markers of group D were singnifcantly increased compared with those of group C （ P 〈0.05 ） ,markers of group S were decreased compaared with those of group D （ P 〈 0.05 ） ; mean glomerular area and total number glomerular cells of group D were higher than those of other groups （ P 〈 0. 05 ） ,expression of vlmentin in group S were decreased compared with those in group D. Conclusion：treatment with simvastatin, the pathology of kidney were improved;simvastatin may play a protective role by downregulating vimentin.
