三七总皂甙对凝血酶诱导VSMCs增殖及pp60c-src活性的影响

摘　　要：目的探讨三七总皂甙(total spaonins of panax notoginseng,tPNS)对凝血酶诱导大鼠血管平滑肌细胞(Vascular smooth muscle cells,VSMCs)增殖作用及与pp60c-src活性的关系。方法采用4-10代的VSMCs的单细胞悬液用于实验。通过MTS法测定VSMCs细胞增殖率。用免疫沉淀测定pp60c-src的活性。结果①0.5U/mL和1.0U/mL2种浓度的凝血酶(Thrombin,TH)促VSMCs增殖相对于对照组,在各时间段均有明显促增殖作用(P<0.05)。增殖曲线呈双峰样改变(峰值在1h和24h);②不同浓度(300.0～600.0μg/mL)的tPNS在24h时对凝血酶刺激VSMCs都有明显抑制作用(P<0.05),且各浓度之间差异有显著性意义(P<0.05)。③相对凝血酶组,tPNS(450.0μg/mL)有效地降低了凝血酶诱导VSMCs增殖的1、24h峰值(P<0.05)。④凝血酶刺激组pp60c-src活性在1h(P<0.01)明显增加,并且6、12、24、48、72h也有增加。用tPNS干预凝血酶诱导VSMCs增殖组,与凝血酶组比较,pp60c-src活性在1h(P<0.01)高峰明显被抑制,并且6、12、24、48、72h也有减少。结论①凝血酶促VSMCs的增殖在时间上呈双峰样改变。②不同浓度的tPNS对凝血酶诱导VSMCs增殖均有抑制作用。③tPNS能抑制凝血酶早期快速及后期促VSMCs增殖作用。④tPNS对凝血酶诱导VSMCs增殖有明显抑制作用,部分机制与其降低pp60c-src活性有关。Objective To study the effect of total spaonins panax notoginseng （tPNS）in thrombin-induced vascular smooth muscle cells （VSMCs） proliferation and pp60c-src activity.Method Growth-arrested VSMCs （4-10 generation） were used in experiment.Cell proliferation was quantified by MTS method.The activity of pp60 c-src in cells was investigated by immunoprecipitation.Results ①Either thrombin group （0.5 or 1.0 U/mL） presents similar cell proliferation curves with two peaks,occurring at 1 h（P0.05） and at 24 h（P0.05）.②tPNS at various concentrations（300.0～600.0 μg/mL） significantly decreased thrombin-induced VSMCs proliferation at 24 h（P0.05）,and the effects of various concentrations of tPNS （300.0～600.0 μg/mL） were different to each other （P0.05）.③Compared with thrombin group,tPNS（450.0 μg/mL）decreased the thrombin-induced cells proliferations at 1 h（P0.05） and at 24 h（P0.05）. ④Compared with 0 h,the activity of pp60c-src in thrombin group increased significantly at 1 h（P0.05）and at 6、12、24、48、72 h respectively.Compared with thrombin group,tPNS decreased the the activity of pp60c-src at 1 h（P0.01）and at 6、12、24、48、72 h respectively.Conclusions ①The cell proliferation induced by thrombin has two peaks at 1 h and 24 h. ②tPNS at various concentrations significantly decreased thrombin-induced VSMCs proliferation.③The inhibition effect of tPNS in thrombin-induced VSMCs proliferation was not only quickly and transient but also torpid and late.④tPNS can inhibit the proliferation of VSMCs induced by thrombin significantly,at least in part by decreaseing the pp60c-src activity.
