摘 要:采用随机扩增多态DNA技术,对不同品种的20只藏獒个体之间的亲缘关系进行了分析。从80条随机引物中筛选出了27条引物,共扩增出369条条带,其中339条表现出多态性,多态性位点百分率为91.87%。根据20只藏獒个体之间的遗传距离,用UPGMA法构建了聚类分析图。结果表明,河曲藏獒和青海藏獒同类群个体之间亲缘关系较近,在聚类图上形成了河曲藏獒类群和青海藏獒类群各自独立的分支,这一结果有力地支持了按地域对藏獒进行类群划分的说法。In this study,the phylogenetic relationship among 20 individuals of Tibetan Mastiff was analyzed by random amplified polymorphic DNA (RAPD) molecular marker technique. Twenty-seven arbitrary primers for RAPD were selected out from 80 primers. A total of 369 bands were amplified,of which 339 were polymorphic,and the percentage of polymorphic RAPDs was 91.86 %. Genetic distance index among 20 individuals of Tibetan Mastiff were calculated and the dendrogram was constructed by UPGMA method. The results showed that the same populations of individuals had close phylogenetic re- lationship between Hequ Tibetan Mastiff and Qinghai Tibetan Mastiff. Separate branch of Hequ Tibetan Mastiff and Qinghai Tibetan Mastiff were formed in the dendrogram, which strongly supports geographical division of the Tibetan Mastiff to taxa argument.
