摘  要:目的了解临床分离大肠埃希菌Ayfy2926中TEM型β内酰胺酶的基因型。方法采用聚合酶链反应(PCR)扩增大肠埃希菌Ayfy2926中TEM型β内酰胺酶的全编码基因并对PCR产物进行序列分析;PCR产物克隆入pHSG398质粒并在感受态细胞大肠埃希菌JM109中进行表达,琼脂稀释法测定转化子菌株对β内酰胺类抗菌药的敏感性。结果大肠埃希菌Ayfy2926中TEM型β内酰胺酶的基因型为一种新亚型,TEM-166(GenBank注册号FJ197316);与TEM-1相比,仅有1个氨基酸差异(Arg120→Gly);与TEM-1转化子菌株相同,TEM-166转化子菌株对哌拉西林高度耐药,对第三代头孢菌素、亚胺培南敏感,β内酰胺酶抑制剂可显著降低克隆表达菌株的耐药性。结论TEM-166为TEM型β内酰胺酶的新亚型,是一种非超广谱β内酰胺酶。Objective To investigate the TEM β-lactamases gene in the isolate of Escherichia coli Ayfy2926.Methods The TEM β-lactamase gene was amplified by PCR and sequenced.The purified PCR product was cloned into pHSG398 vector and transformed into E.coli JM109.The antibiotic susceptibility of transformant was tested by agar dilution method.Results The TEM β-lactamases gene was identified as a new subtype,TEM-166,with 1 amino acid substitution between TEM-1 and TEM-166(GenBank accession no.FJ197316).The transformant with TEM-166 gene was resistant to piperacillin and susceptible to the third generation cephalosporins,imipenem and piperacillin-clavulanic acid.Conclusions TEM-166 is a new subtype of TEM β-lactamase,which is belonged to non-extended spectrum β-lactamase.
