摘 要:目的使用虚拟的电压信号发生器,以及与电极、计算机的组合技术,探讨并建立细菌氧化反应的实验模型。方法虚拟的电压信号发生器由LabVIEW软件、多功能数据采集卡及计算机组成。计算机控制虚拟的电压信号发生器,输出阶梯形递增电压(0.00~1.00V,递增速率:10mV/s),加到电极上。电极系统由平面石墨电极、铂电极及饱和甘汞电极组成。使用大肠埃希氏菌,把附着细菌的纸质滤膜固定在工作电极表面,在工作电极与对极之间施加阶梯形递增电压,计算机同步记录伏安图谱。通过伏安图谱分析电极间的电子流动变化,探讨电子流动与细菌氧化反应之间的关联。结果纸质滤膜上附着大肠埃希氏菌时,伏安图谱显示施加电位0.72V(vs.SCE)附近出现电流的峰谷,峰电流值随菌浓度增大相应增高。纸质滤膜上未附着大肠埃希氏菌时,伏安图谱未显示出电流的峰谷。结论施加电位0.72V(vs.SCE)附近出现电流的峰谷,为大肠埃希氏菌氧化反应所释放的电子形成。大肠埃希氏菌的氧化反应,与菌体内辅酶A相关。Objective Virtual voltage signal generator, combined with electrodes and computer technology were used to explore and establish the experimental model of bacterial oxidation reaction. Methods The virtual voltage signal generator consists of the Lab VIEW software, muhifunctional data acquisition card and computer. Computer controls the virtual voltage signal generator, outputs ladder-shaped incremental vohage (0.00 ~ 1.00 V, scan rate: 10 mV/s) to add on the electrodes. Electrode system consists of basal plane pyrolytic graphite, platinum electrode and saturated calomel electrode. Escherichia coli was fixed on the bacteria paper upon the working electrode surface, ladder-shaped incremental voltage between the working electrode and counterelectrode was exerted, and the computer simultaneously recorded vohampere map. The electronic flow changes between the electrodes were analyzed to explore the association of electronic flow and bacterial oxidation reaction. Results The voltampere map shows current peak in the vicinity exerted voltage 0.72 V ( vs. SCE) when membrane is attached by Escherichia coli, and the current peak increases with the concentration of bacteria. When the membrane has no attaehed Escherichia coli, the vohampere map does not show current peak. Conclusion The current peak in the vicinity exerted voltage 0.72 V (vs. SCE) is due to the electrons released by the oxidation of Escheriehia coll. The oxidation reaction of Eseherichia eoli was associated with CoA of bacteria in vivo.
