慢性HBV感染者外周血T细胞CD127的表达与IL-7及HBV载量的相关性

摘　　要：目的通过检测HBV感染者外周血T淋巴细胞表面CD127(IL-7受体)的表达,探讨其与HBV载量及疾病进程的相关性。方法选取125名HBV感染者及40名健康人对照,用流式细胞术检测外周血T细胞表面CD127的表达,ELISA法检测血浆IL-7浓度,实时荧光定量PCR法检测HBV DNA并进行分析。结果HBV感染者外周血CD4+、CD8+T淋巴细胞表面CD127的表达分别为(84.59±5.69)%和(38.45±3.03)%,其中CD8+T淋巴细胞表面CD127的表达明显低于健康对照组(P<0.01);慢性乙肝组外周血CD8+T淋巴细胞表面CD127的表达明显高于慢性重型乙肝组(P<0.05);HBV感染者血浆中IL-7水平明显高于健康对照组,且与CD8+T淋巴细胞表面CD127的表达呈负相关(r=-0.341,P<0.05);HBV病毒载量的对数值与外周血T淋巴细胞表面CD127的表达之间存在负相关(r=-0.432,P<0.01)。结论HBV感染者外周血T淋巴细胞表面CD127的表达水平与疾病进展明显相关。Objective To investigate the correlation between the progression of HBV infection and the IL-7 receptor on T lymphocytes by detecting the expression of CD127 on the surface of T cells in HBV infected patients.Method The expression of CD127 on the surface of T lymphocytes from 125 HBV infected patients and 40 healthy controls was detected by flow-cytometry.Serum IL-7 level was measured by ELISA.HBV DNA was quantitatively detected with real-time PCR.Results The positive rates of CD127 on CD4 and CD8 T cells in HBV infected patients were（84.59±5.69）% and（38.45±3.03）% respectively.The expression of CD127 on CD8 T cells in the patients decreased significantly（P〈0.01） compared with that in healthy controls.The expression of CD127 in the patients was higher than that in the severe patients（P〈0.05）.The serum level of IL-7 in the patients was higher than that in healthy controls,and was negatively correlated with the expression rate of CD127 on the CD8 T cells（r=-0.358,P〈0.05）.The HBV load was negatively correlated with the expression rate of CD127 on CD8 T cells（r=-0.432,P〈0.05）.Conclusions The expression of CD127 on CD8 T lymphocyte is closely correlated with the progression of HBV infection.
