摘 要:目的研究Mu受体在小鼠树突状细胞中的诱导表达。方法取7~8周龄的C57BL/6J雌鼠骨髓细胞培养,培养7d后,用CD11c+磁珠,采用磁性分选的方法纯化树突状细胞。用免疫荧光染色PCR检测Mu受体及其基因在树突状细胞中的表达。并用流式细胞仪对树突状细胞的表型进行分析。结果Mu受体在激活的树突状细胞上有明显(P<0.05)表达。不同Toll样受体的配体诱导不同程度的Mu受体表达。结论活化的树突状细胞可诱导表达Mu受体。Objective To investigate whether murine dendritic cells(DCs)express Mu-opioid receptors.Methods Bone marrow cells were flushed from the femurs and tibiae of normal female,7-to 8-wk-old C57BL/6J mice.After 7 days of culture,CD11c+ DCs were purified by immunomagnetic sorting using anti-CD11c-coated magnetic beads.Gene expression was detected by PCR.Mu-opioid expression was analyzed by double-immunofluorescence staining.DC surface molecules were detected by Florescence-activated cell sorting(FACS).Results The expression of Mu-opioid receptor occurred significant(P0.05)in activated DCs.Different Toll-like receptor's ligands could induce Mu-opioid receptor expression in murine DCs discriminately.Conclusion The results suggest that Mu-opioid receptor is expressed inducibly on activated DCs.
