摘  要:目的探讨沙苑子黄酮(FAC)对CCl4致小鼠慢性肝纤维化的保护作用。方法制备小鼠慢性肝纤维化模型,给予FAC灌服,并以秋水仙碱为对照,检测血清ALT、AST、ALB及肝组织Hyp、MDA、SOD水平的变化。同时观察肝组织的病理变化和肝组织中过氧化物酶体增殖物激活受体γ蛋白的表达情况。结果与正常对照组相比,模型组小鼠血清ALT、AST水平显著升高,ALB水平显著降低(P<0.01)。与模型组相比,FAC各剂量组小鼠血清ALT、AST显著降低,ALB水平显著升高(均P<0.01)。FAC各剂量组可显著升高肝组织中SOD活性及PPARγ的表达,降低MDA、Hyp的含量(P<0.01或P<0.05)。结论FAC有显著改善肝功能、预防肝纤维化作用,其机制可能与其抗脂质过氧化和提高机体的抗氧化酶活性有关。Objective To observe the protection of flavonoids from astragalus complanatus(FAC)on hepatic fibrosis induced by carbon tetrachloride(CCl4)in mice.Methods Copy the model of mice with chronic liver injury induced by carbon tetrachloride.In the meantime,fill mice stomach with the different dose of FAC,comparing with the colchicine pills on a daily basis for four weeks.Observe the serum alanine amino transferase(ALT),aspartate amino transferase(AST)activity,the albumin(ALB)levels were determined biochemically;Liver samples were also analyzed malondiadehyde(MDA)level,superoxide dismutase(SOD)activity and the hydroxypmline(Hyp)level.The damage of liver tissues was observed by light microscope.The expression of peroxisome proliferator-activated receptors γ(PPARγ)were detected by immunohistochemistry.Results CCl4 injection resulted in an increase of the ALT and AST levels in serum,a decrease of ALB levels in serum.FAC treatments reduced the ALT and AST levels,and restored the ALB contents compared with the model group(P0.01).CCl4 injection led to dramatic decrease in SOD activities and increase in MDA and HYP contents.FAC treatments changed their levels(P0.01 or P0.05).The expression of PPARγ decreased during the development of fibrosis.In contrast,the PPARγ positive expression in FAC treated mice remarkably increased compared with the model group(P0.01).Conclusion FAC play a very important role in the improvement of liver function and anti-liver injury among these groups which was related with scavenging free radical,increasing the activities of antioxidant enzyme.
