PTEN基因剪接变异体与肝细胞肝癌发生的相关研究

摘  要:目的研究PTEN基因在肝细胞肝癌中是否存在新的剪接变异体,探讨PTEN基因在肝细胞肝癌发生发展中的作用。方法提取肝细胞肝癌组织的RNA,逆转成cDNA。根据Genbank上的PTEN基因cDNA序列,设计6对引物,并以肝细胞肝癌组织cDNA为模板分别扩增外显子1~2、1~3、2~4、3~5、4~6和5~7之间的片段,得到的PCR产物进行3%琼脂糖凝胶电泳,比较不同样本扩增出来的目的条带的异同。结果每对引物分别做了61例肝细胞肝癌组织,3%琼脂糖凝胶电泳后发现每对引物扩出来的目的条带大小相同,与野生型剪切体一致。结论PTEN基因在肝细胞肝癌中不存在新的剪接变异体,说明在肝细胞肝癌发生发展中PTEN没有出现异常的mR-NA剪接突变。Objective To screen for novel splicing variants of PTEN gene in hepatocellular carcinoma(HCC)patients and discuss its significance during the development of HCC.Methods RNA was exacted from HCC tumor tissues,and was reversely transcribed into cDNA.According to PTEN sequences from Genbank,six pairs of different primers were designed to amplify exon 1 to exon 2(1~2),exon 1 to exon 3(1~3),exon 2 to exon 4(2~4),exon 3 to exon 5(3~5),exon 4 to exon 6(4~6)and exon 5 to exon 7(5~7)using the cDNA template.Three percent agarose gel electrophoresis was used to detect PCR products,and the sizes of the products were compared.Results Sixty one HCC patient samples were screened with each pair of primers;the amplified PCR products exhibited the same sizes as the wild type PTEN transcripts.Conclusion Novel splicing variants of PTEN gene are not found in HCC tumor samples,which suggests splicing mutant of PTEN may be present in HCC.
