摘 要:目的探讨IL-1β、TNF-α及TGF-β1在染尘矽肺大鼠肺组织内的变化和对Ⅰ、Ⅲ型胶原含量的影响。方法将50只大鼠随机分为对照组、染尘组,各组于1、7、14、21、28d5个时间点分别取5只大鼠,采用气管暴露法建立大鼠矽肺模型;分别进行HE常规染色、天狼猩红染色并用Image-pro Plus Version 4.5 for WindowsTM图像分析系统进行Ⅰ、Ⅲ型胶原定量分析;肺组织内IL-1β、TNF-α、TGF-β1测定采用悬浮芯片蛋白测定法。结果Ⅰ、Ⅲ型胶原分别在第14天和第7天开始急剧增高;染尘组肺组织IL-1β的浓度在第1天达到高峰;TNF-α的表达量持续增加,在第14天出现峰值;TGF-β1第14天表达量增加并达峰值;回归分析结果显示IL-1β表达水平下降和TNF-α表达水平升高可能直接或间接导致Ⅰ型胶原的增加;IL-1β表达水平下降、TGF-β1表达水平升高可能直接或间接导致Ⅲ型胶原的增加。结论矽尘可以诱导肺组织过度表达Ⅰ、Ⅲ型胶原,并且IL-1β、TNF-α及TGF-β1可以直接或间接地增加胶原含量。Objectives To explore the relationship between contents of collagen Ⅰ or Ⅲ and levels of IL-1β, TNF-α or TGF-β1 in rats exposed to silica.Methods Rats were exposed to 50 mg silica suspended in 0.9% saline solution through intratracheal instillation, then were sacrificed at 1, 7, 14, 21 and 28d after a single instillation, respectively. Lungs were removed and stained with hematoxylin and eosin (HE) for pathological examination or with Sirius red stains for determination of collagen Ⅰ and Ⅲ contents. Levels of IL-1β,TNF-α and TGF-β1 in lung were determined by suspension array technique. Results Compared with those in control group, the contents of collagen Ⅰ were increased on the 14th day after exposure, and the contents of collagen Ⅲ were on the 7 th day. The levels of IL-1β were increased significantly on the 1st day; levels of TNF-α were increased on the 14 th day, then decreased on the 28th day. The levels of TGF-β1 were increased significantly between the 14th day and 28th day. Correlation analysis showed that the levels of IL-1β and TNF-α were correlated with the content of collagen Ⅰ, and the levels of IL-1β and TGF-β1 were also correlated significantly with content of collagen Ⅲ. Conclusions The results showed that the silica dust might induce the overexpression of collagen Ⅰ or Ⅲ, IL-1β, TNF-α and TGF-β1, and the latter three indices might play the important role in the formation of collagen in the lung.
