摘 要:目的:探讨镍离子(Ni2+)、六价铬离子(Cr6+)联合染毒对大鼠睾丸波形蛋白表达的影响。方法:40只雄性SD成年大鼠随机分为10组,每组4只。实验试剂硫酸镍(NiSO4)和重铬酸钾(K2Cr2O7)以质量分数为0.9%NaCl溶解,通过腹腔注射给药,连续染毒2周。A^J组每天用药的质量分数分别为:A组:不作任何处理;B组:0.9%NaCl溶液;C组:1.25 mg/kg Ni2+;D组:5 mg/kg Ni2+;E组:2 mg/kg Cr6+;F组:3 mg/kg Cr6+;G组:4 mg/kgCr6+;H组:1.25 mg/kg Ni2++2 mg/kg Cr6+;I组:5 mg/kg Ni2++3 mg/kg Cr6+;J组:5 mg/kg Ni2++4 mg/kg Cr6+。检测大鼠体质量、睾丸湿质量及睾丸脏器系数的变化。应用免疫组织化学方法检测睾丸波形蛋白的表达和变化。结果:E^J组大鼠体质量和睾丸湿质量与对照组相比较均显著降低(P<0.01),各实验组睾丸脏器系数无显著变化(P>0.05)。C^J组支持细胞及间质区波形蛋白表达量与对照组相比较均显著降低(P<0.01)。H组与C、E组相比较,I组与D、F组相比较,J组与D、G组相比较,其支持细胞和间质区波形蛋白的表达量均显著降低(P<0.01)。结论:Ni2+、Cr6+染毒能显著降低大鼠体质量、睾丸湿质量以及睾丸波形蛋白的表达,Ni2+-Cr6+联合染毒对睾丸损伤更严重。Aim:To investigate the combined effects of nickel(Ni^2+) and hexavalent chromium(Cr^6+) on vimentin expression in adult rat testes. Methods: Forty male adult SD rats were divided into 10 groups randomly.Rats were administrated by intraperitoneal injection of various chemicals for 2 weeks.NiSO4 and K2Cr2O7 were solved in normal saline and given at the dose of group A: no treatment,group B: 0.9% NaCl,group C: 1.25 mg/kg·day Ni^2+,group D: 5 mg/kg·day Ni^2+,group E: 2 mg/kg·day Cr^6+,group F: 3 mg/kg·day Cr^6+,group G: 4 mg/kg·day Cr^6+,group H: 1.25 mg/kg·day Ni^2++2 mg/kg·day Cr^6+,group I: 5 mg/kg·day Ni^2++3 mg/kg·day Cr^6+ and group J: 5 mg/kg·day Ni^2++4 mg/kg·day Cr^6+.Testes wet weight,body weight and their ratio were analysed.Immunohistochemistry method was used to determine the expression of vimentin in testes.Results: Compared with control,testes wet weights and body weights in groups E-J all decreased significantly(P〈0.01).There was no significant differences in ratios of testes wet weights to body weights in each experimental group(P〉0.05).Expression of vimentin in sertoli cells and testicular interstitium of groups C-J were all decreased significantly compared with control(P〈0.01).Furthermore,the decrementin group H,I and J was larger than that in decrease compared with group C or E,group D or F and group D or G,respectively(P〈0.01).Conclusion:Exposure of Ni^2+ or Cr^6+ would decrease the body weight,testes wet weight and the expression of vimentin.The combination of Ni^2+ and Cr^6+ could cause more damage to testes.
