摘  要:目的优化黄精多糖的提取纯化工艺及建立其薄层鉴别方法。方法以多糖提取率为指标,采用正交实验设计,优选复方中黄精多糖的提取纯化工艺;采用多展开剂系统筛选黄精的薄层鉴别方法。结果黄精多糖最佳水提工艺为:8倍量水,提取2次,每次1h;最佳纯化工艺为药材:浸膏比为1:0.7;用80%乙醇醇沉,醇沉1次。主药黄精的最佳薄层展开剂条件为氯仿:丙酮(4:1)。结论黄精多糖的提取纯化工艺稳定、操作简单、切实可行;主药的薄层色谱条件分离效果、重现性好。Objective To select the optimum extraction and purification process for polysaccharide from rhizoma polygonati,and establish TLC identification method.Methods The optimum extraction and purification process was selected with the content of rhizoma polygonati by orthogonal test design.TLC identification method of rhizoma polygonati is screened by the methods of multiple solvent system and elimination the negative interference.Results The optimum extraction and purification process was determined follows as,adding 8 times of water decocting twice for 1 h each time,and alcohol precipitation once by ethanol of 70%,the best solvent system was chloroform:acetone=4:1(volume ratio).Conclusion The extraction and purification process is reasonable,stable and favorable for industrial production.The conditions of thin-layer chromatography has good separation effect and reproducibility.
