摘 要:目的:研究复方SZ滴眼液(Compound shui zhi eye drop,SZ)和阿魏酸钠(Sodium ferulate,SF)对紫外线(Ultraviolet,UV)损伤的人晶状体上皮细胞抗氧化酶及其基因表达的影响。方法:采用UV照射体外培养的人晶状体上皮细胞(Human lens epithelial cell,HLEC),同时用SZ、SF和吡喏克辛钠滴眼液(Pirenoxine sodium,PS)与HLEC共同孵育,并检测HLEC中超氧化物歧化酶(Superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(Glutathi-one peroxidase,GSH-px)和过氧化氢酶(Catalase,CAT)的活性和SODmRNA、GSH-pxmRNA和CATmRNA的基因表达。结果:UV组的HLEC抗氧化酶活性和基因表达均比对照组显著降低(P<0.01)。SZ、SF和PS组的SOD、GSH-px和CAT的活性显著高于UV组(P<0.01,P<0.05);SODmRNA、GSH-pxmRNA和CATmRNA的表达也显著高于UV组(P<0.01)。结论:SZ、SF和PS均能防护UV照射引起的HLEC氧化损伤,SZ和SF的作用优于PS。Objective:To investigate the effect of compound shui zhi eye drop (SZ) and sodium ferulate (SF)on anti-oxidase and its gene expression in human lens epithelial cell(HLEC) injured by ultraviolet(UV).Methods:HLEC were irradiated by UV.The HLEC were incubated with SZ,SF and pirenoxine sodium(PS).The activities of superoxide dismutase (SOD).glutathione peroxidase (GSH-px ) and catalase (CAT) were measured and the gene expression of SODmRNA,GSH-pxmRNA and CATmRNA were detected.Results:The activities of anti-oxidase and its gene expression of HLEC in UV group decreased significantly compared with control group (P〈0.01).The activities of SOD,GSH-px and CAT in SZ group,SF group and PS group were higher than that in UV group significantly (P〈0.01,P〈0.05).The gene expression of SODmRNA、GSH-pxmRNA and CATmRNA in SZ group,SF group and PS group were higher than that in UV group remarkably (P〈0.01).Conclusion:SZ,SF ang PS can prevent and protect oxidative damages in HLEC injured by UV;The effects of SZ and SF are better than PS.
