摘 要:目的:观察熊果酸对高脂饮食诱导的胰岛素抵抗(Insulin resistance,IR)大鼠脂质代谢及IR的作用。方法:采用高脂饲料持续喂养建立IR大鼠模型,熊果酸灌胃给药进行干预,二甲双胍作为阳性对照药物,分别在给药第4、8周后,测定体重、空腹血糖(fasting blood glucose,FBG)、空腹血清胰岛素(fasting serum insulin,FINS)、血清总胆固醇(TC)、三酰甘油(TG)、游离脂肪酸(free fatty acids,FFA)、肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)、肝脏、腹腔脂肪重量,计算肝指数(liver index,LI)、腹脂指数(adipose index,AI)、胰岛素敏感性指数(insulin sensitive index,ISI)和胰岛素抵抗指数(HOMA-IR),评价熊果酸对IR大鼠脂质代谢的影响。结果:熊果酸能够显著降低IR大鼠血清FINS、TC、TG、FFA和TNF-α水平,减少腹腔脂肪堆积,增加胰岛素敏感性,与模型组比较差异有统计学意义(P<0.05或P<0.01)。结论:高脂饮食可导致大鼠胰岛素抵抗,熊果酸能够调节脂质代谢、改善胰岛素抵抗。Objective:To investigate the effects of ursolic acid on fat metabolism and insulin resistance induced by high fat diet in rats.Methods:Insulin resistant rats were induced by high fat diets,then the rats were intervened by ursolic acid or metformin through intragastric administration respectively.Levels of body weight (BW),fasting blood glucose (FBG),fasting serum insulin (FINS),TC,TG,free fatty acids (FFA),tumor necrosis factor-α (TNF-α),liver weight (LW) and abdominal cavity adipose weight (AW) were determined following 4 weeks or 8 weeks administration,and the liver index (LI),adipose index (AI),insulin sensitive index (ISI) and HOMA-IR were calculated to evaluate the effects of ursolic acid.Results:Ursolic acid could markedly reduce the levels of FINS,serum TC,TG,FFA,TNF-α and the adipose weight of abdominal cavity,and improve the insulin sensitivity in IR rats,as compared with those of untreated rats (P〈0.05 or P〈0.01).Conclusion:High fat diet resulted in insulin resistance.Ursolic acid could improve insulin resistance and fat metabolism.
