保肝汤对大鼠酒精性肝损伤治疗的实验研究

摘　　要：目的:探讨保肝汤对酒精性肝损伤的治疗作用,并探讨作用机理。方法:56只昆明种小鼠随机分为正常组、模型组、保肝汤低剂量组、保肝汤高剂量组,每组14只。采用白酒灌胃造成酒精性肝损伤模型,造模同时行药物治疗,6周后处死动物后进行肝脏组织病理学检查观察。取血用分光光度仪检测血清肝功能酶AST、ALT的变化,免疫组化方法检测基质金属蛋白酶-13(MMP-13)和基质金属蛋白酶抑制剂-1(TIMP-1)。初步探讨其对酒精性肝损伤的作用机理。结果:模型组肝小叶结构紊乱,大量气球样变及脂肪变性。保肝汤低剂量组汇管区及中央静脉周围胶原纤维组织增生较模型组明显减少。同模型组和保肝汤高剂量组相比具有明显差异。保肝汤可以明显降低ALT、AST的含量,与模型组比较具有显著差异(P<0.05)。保肝汤组肝组织MMP-13蛋白的表达均较模型组有明显的增强(P<0.05),保肝汤组肝组织TIMP-1蛋白的表达均较模型组有明显的减弱(P<0.05)。结论:保肝汤可以通过抑制纤维化对酒精性肝损伤具有治疗作用。Objective：Study the protection of baogan decoction to alcoholic liver injury.Methods：56 kunming mice were randomly divided into normal group,the model group,low dose group of baogan decoction,high-dosage groups of baogan decoction,Each group of 14.Gastric lavage caused by liquor alcoholic liver damage model,at the same drug therapy,After six weeks,animal＇s liver histopathological examination.Take blood serum with spectrophotometric detection AST and ALT.Immunohistochemical method detection MMP-13 and TIMP-1.Preliminary discussion on the alcoholic liver damage mechanism.Results：the model group lobular architecture disorder,a large number of ballooning degeneration and fatty degeneration.low dose group of baogan decoction of collect abbacy and central venous surrounding collagenous fiber hyperplasia in model group significantly reduced.Compared with the model group and baogan decoction group were obvious difference.Baogan decoction group can obviously reduce ALT and AST,compared with model group were significant difference （P〈0.05）.MMP-13 protein of liver tissue of baogan decoction group expression was significantly enhanced the model group （P〈0.05）,TIMP-1 protein liver tissue of baogan decoction group expression as significantly weakened,compared with model group （P〈0.05）.Conclusion：Baogan decoction has protection to alcoholic liver fibrosis.To baogan decoction group can inhibit fibrosis of alcoholic liver injury therapeutic effect.
