摘 要:[目的]探讨上游转录因子1(upstream transcription factor1,USF1)基因多态性rs3737787与糖脂代谢相关指标的关系。[方法]采用以核心家系为基础的连锁与关联研究设计,应用PCR-RFLP方法对287个2型糖尿病核心家系成员USF1基因rs3737787多态性进行分析。[结果]在非参数连锁分析中,rs3737787多态性与血浆甘油三酯与载脂蛋白E水平有连锁关系,其LOD(logarithm of odds)值分别为0.87(P=0.02)与1.96(P=0.001)。应用广义估计方程(generalized estimate equation,GEE)控制其他混杂因素并调整家系内部相关性后,rs3737787多态性与这两种性状的关联仍具有统计学显著性。[结论]研究结果提示USF1基因rs3737787多态性与2型糖尿病家系人群的血浆甘油三酯及载脂蛋白E水平相关。[Objective] To explore the association between rs3737787 in upstream transcription factor 1 (USF1) and glucose and lipid metabolic profile. [Methods] A total of 287 eligible T2DM families were chosen. DNA was extracted from blood samples and genotyped using PCR-RFLP method. Statistical analyses including linkage analysis and family-based association test were performed to detect the relationship between rs3737787 and traits of blood glucose and lipid profile. [Results] In the non-parametric linkage analysis, it was observed that rs3737787 may have potential linkage relationship with the triglyceride and apoprotein E traits, while the logarithm of the odds were 0.87 (P = 0.02) and 1.96 (P = 0.001), respectively. Similar results were obtained in the multi-factorial generalized estimating equation analysis. [Conclusion] In this study, we have confirmed that the single nucleotide polymorphism rs3737787 is related with triglyceride and apoprotein E level in type 2 diabetes mellitus families.
