摘 要:为验证对黄萎病菌(Verticllium daliae)、枯萎病菌(Fusarium oxysporum f.sp.vasinfectum)的早期抗性反应,确定了转hpa1Xoo基因棉花株系T-33悬浮细胞适宜的培养条件,并成功获得了T-33和其出发品种Z35的悬浮细胞。以1:20的比例,将T-33悬浮细胞与黄、枯萎菌分生孢子液分别共培养0~24h,发现T-33悬浮细胞在0~6h对两种病原菌的抗性能力较强,悬浮细胞死亡率显著低于Z35(P<0.05〉。与Z35相比,在3h和6h时,针对棉花黄萎病菌T-33悬浮细胞死亡率分别低52%和35%,针对枯萎病菌分别低42%和25%。说明hpa1Xoo编码的harpinXoo表达赋予转基因棉花T-33悬浮细胞系对两种病原菌的抗性高于其出发品种Z35。For confirming the earlier resistance reaction,the suspension cells of transgenic hpa1Xoo cotton plant T-33 and untransformed cotton plant Z35 were cultured successfully,and the suspension cell of T-33 plant were mixed and co-cultured with Verticllium daliae and Fusarium oxysporum f.sp.vasinfectum conidia at 1:20 ratio for 0~24 h respectively.Z35 was a control.The death rate of the suspension cell of T-33 was significantly lower than that of Z35 during 3~12 h culture time,in particular reduced 52% at 3 h and 35% at 6 h to V.dahliae,and reduced 42% at 3 h and 25% at 6 h to F.oxysporum f.sp.vasinfectum.The evidence indicated the expression of harpinXoo encoded by hpa1Xoo enhanced the resistance of T-33 suspension cells to two pathogens compared with Z35 suspension cells.
