摘 要:利用限制性培养技术,构建了1组35℃下遗传稳定的、产甲烷量高的沼气发酵复合菌系。采用单因素及正交试验分析方法,研究了pH、接种量、底物浓度三个因素的培养条件对沼气复合菌系甲烷产率的影响。试验结果表明,在单一因素的影响下,发酵初始的pH值为7.0,接种量为10%,底物浓度为8%时产甲烷效果最好。采用正交试验进行培养条件的优化组合试验分析,确定最佳的沼气复合菌系发酵条件为:以牛粪为底物,复合菌系接种量5%,底物浓度为10%,pH自然,培养温度为35℃。A microbial community capability of biogas production with genetic stability at 35 ℃ was enriched from biogas slurry with the method of limited culture technique,and the optimum cultural conditions of microbial community were investigated with single factor analysis and orthogonal test analysis.The effect of different pH,inoculation quantities,substrate concentrations of microbial community were investigated.The results of single factor design indicated that the optimum cultural conditions of microbial community for biogas production were the pH value 7.0,10% of inoculation quantity,and 8% of substrate concentration,respectively.At the same time,the results of orthogonal test analysis showed that the optimum cultural conditions of microbial community for biogas production using the cattle manure as material were 5% of inoculation quantity,10% of concentration of substrate,natural pH and temperature 35 ℃,respectively.
