摘 要:目的探讨肾乐胶囊对慢性肾炎兔及慢性肾功能衰竭(肾衰)大鼠的治疗作用。方法利用兔慢性血清病肾炎模型、大鼠5/6肾切除肾衰模型、大鼠血瘀模型及小鼠水负荷模型,每个模型实验分为6组:正常对照组(假手术组),模型对照组,药物对照组,肾乐高、中、低剂量组。测定慢性血清病肾炎模型兔肾功能及24 h尿蛋白定量,ELISA法测定血清循环免疫复合物(CIC),MTT掺入法测定脾细胞白细胞介素2(IL-2)活性。测定5/6肾切除肾衰模型大鼠肾功能及24 h尿蛋白定量,并观察其肾脏病理改变。测定血瘀模型大鼠血液流变学指标及水负荷小鼠排尿量。结果与模型对照组比较,肾乐各剂量组给药6 wk血肌酐、尿素氮,24 h尿蛋白量降低(P<0.05),肾脏病理损害减轻,血清(白)蛋白升高(P<0.01),CIC和脾细胞IL-2活性下降(P<0.05)。肾乐各剂量组大鼠血液黏滞度均显著降低(P<0.01),水负荷小鼠给药后排尿量增加。结论肾乐胶囊能减轻肾小球炎症损害,改善肾功能,对慢性肾炎和慢性肾衰有一定疗效。AIM To study models and renal failure rats. the therapeutic effects of Shenle capsule (SLC) on chronic nephritis rabbit METHODS The experimental animal models with chronic serum sickness nephritis rabbits, blood stasis model rats, water load model mouse and 5/6 nephrectomy rats were used. The experimental animals were divided into 6 groups: normal group (sham group), model group, high-dose SLC group, moderate-dose SLC group, low-dose SLC group and drug control group. Serum creatinine (Scr), urea nitrogen (BUN), albumin (Alb) and urine protein were monitored at different time point. ELISA was used to examine the content of circulating immune complex (CIC) . The activity of IL-2 was measured by MTT assay. Renal biopsy was performed to study the pathology changes. Hemorheology in blood stasis model rats and the urine output of water load mice were also measured. RESULTS Compared with the model group, in the SLC groups, urine protein, Scr, BUN decreased (P 〈 0.05) , renal pathological lesions were lightened (P 〈0.01) , and blood Alb was synthesized (P 〈 0.01) significantly decreased in the SLC groups (P 〈 0.05) . The content of CIC and the activity of IL-2 were And SLC had the function of diuresis and invigorating the circulation of blood. CONCLUSION SLC can reduce glomerular inflammation and protect renal function, and has some therapeutic effects on chronic nephritis and renal failure.
