摘  要:目的探讨不同浓度葛根素对大鼠肾小球系膜细胞增殖的影响及其作用机制。方法采用体外细胞培养技术,采用MTT检测不同浓度葛根素对大鼠肾小球系膜细胞增殖的量效和时效关系。分别采用RT-PCR和ELISA,检测不同浓度葛根素作用大鼠肾小球系膜细胞后,结缔组织生长因子mRNA和蛋白表达水平的变化。结果葛根素可抑制大鼠GMCs增殖(P<0.05),该增殖抑制作用呈浓度依赖模式。不同浓度葛根素作用于大鼠GMCs后,结缔组织生长因子mRNA和蛋白表达水平均随药物浓度增加而逐渐减弱(P<0.05)。结论葛根素对大鼠肾小球系膜细胞增殖有明显的抑制作用,其作用机制可能与下调结缔组织生长因子基因转录和表达水平有关。OBJECTIVE To determine the effect and mechanism of puerarin on proliferation of rat glomerular mesangial cells(GMCs).METHODS The dosage-and time-effect relationships among different concentrations of puerarin and proliferation of GMCs were investigated by cell culture in vitro using MTT.RT-PCR and ELISA were applied to measure the mRNA and protein expression levels of connective tissue growth factor(CTGF) in GMCs treated with different concentrations of puerarin,respectively.RESULTS The result of MTT showed that puerarin inhibited the proliferation of GMCs with a dosage-dependent manner(P〈0.05).The results of RT-PCR and ELISA indicated that the mRNA and protein expression levels of CTGF in puerarin-treated GMCs were gradually decreased following the increase of puerarin dosages(P〈0.05).CONCLUSION Puerarin possesses an obvious inhibitory effect on proliferaton of GMCs and the action mechanism may refer to down-regulation of transcription and expression levels of CTGF encoding gene.
